ICY1 has synthetic growth impaired phenotypes with tcp1-1 and with the ICY2 null mutant. Knock out affects cell shape, nuclear morphology and movement and membranous vesicle trafficking. Overexpression partially suppresses tcp1-2. Null mutant is hypersensitive to rapamycin (less than icy2 null), and the hypersensitivity is suppressed by the tcp1-1 mutation. The null mutant has defects in nuclear transport. The icy1 icy2 double null is conditionally lethal at 15 deg C, hypersensitive to latrunculin A, displays defects in chromatin organization and nuclear transport, and is super-sensitive to alpha factor, using a disk assay. The double mutant arrests for growth in two stages, with a characteristic double halo. In addition, vacuole morphology of the double null mutant is aberrant, exhibiting a larger number of vacuole lobes than either single mutants or the wild-type.
